| REG1a

Pancreatic Stone Protein (PSP), also known as Lithostathine-1-alpha islet cells regeneration factor (ICRF) or islet of Langerhans regenerating protein (REG) is a protein that is encoded by the REG1A gene as a single polypeptide of 144 amino acids further cleaved by trypsin to produce a 133 amino acid protein that is O-linked glycosylated on threonine 27. This protein is a type I subclass member of the Regenerating protein family. The PSP has been shown to be associated with islet cell regeneration and diabetogenesis and may be involved in pancreatic lithogenesis (creation of pancreatic stones; pancreatic calculi).

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (pg/mL) 4000 2000 1000 500 250 125 62.5 0
OD 2.21 1.48 1.03 0.69 0.41 0.3 0.19 0.05
Corrected OD 2.16 1.43 0.98 0.64 0.36 0.25 0.14 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human REG1a spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-101 96
Plasma(n=8) 92-105 102

| Linearity

Samples were spiked with high concentrations of Human REG1a and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4)
1 : 2 Range (%) 85-95 88-96
Average (%) 91 93
1 : 4 Range (%) 89-103 87-108
Average (%) 94 98
